Symbiodinium isolation by NaOH treatment.

چکیده
The presence of photosynthetic zooxanthellae (dinoflagellates) in the tissue of many cnidarians is the main reason for their ecological success (i.e. coral reefs). It could also be the main cause of their demise, as the worldwide bleaching of reef-building coral is nothing less than the breakdown of this symbiotic association. The stability of this relationship is the principal marker for the biomonitoring of cnidarian health. We have therefore developed a new, simple method to isolate zooxanthellae in a few steps using NaOH solution. The protocol was validated in three symbiotic cnidarian species: a sea anemone, a gorgonian and a coral. Our method allows the isolation of intact and viable zooxanthellae with better yields than classic methods, especially for species with a calcareous skeleton. Moreover, the isolated zooxanthellae were free of host nucleic contaminants, facilitating subsequent specific molecular analyses.

Phylogenetic Analysis of a Free-living Strain of Symbiodinium Isolated from Jiaozhou Bay, P.r. China
Recently, the isolation of a free-living strain of symbiotic dinoflagellate belonging to the genus Symbiodinium was reported. Although the specimen procured and characterization from Jiaozhou Bay, P.R. China is a Symbiodinium spp., the manner in which this isolate was classified is inconsistent with the currently used and accepted Symbiodinium cladal nomenclature. To avert unnecessary confusion...
متن کاملOptimized Method for Isolation of Nontuberculous Mycobacteria from Hospital Aquatic Sources
Background: Aquatic ecosystems are an important source of nontuberculous mycobacteria (NTM) that can cause different diseases in human. Since culture of mycobacteria needs long-term incubation, fast-growing microorganisms and contaminants in the environment usually prevents the isolation of mycobacteria. Here, we compare different treatment protocols and describe a method that increases the rec...
متن کاملRecovery of mycobacteria from patients with cystic fibrosis.
Despite decontamination, overgrowth by pseudomonads renders cultural isolation of mycobacteria from respiratory specimens of patients with cystic fibrosis (CF) difficult or impossible. We performed a prospective study by comparing levels of reduction of overgrowth and recovery of mycobacteria using either pretreatment with N-acetyl-L-cysteine (NALC)-NaOH alone or pretreatment with NALC-NaOH and...
متن کاملChemical decontamination with N-acetyl-L-cysteine-sodium hydroxide improves recovery of viable Mycobacterium avium subsp. paratuberculosis organisms from cultured milk.
Mycobacterium avium subsp. paratuberculosis is shed into the milk and feces of cows with advanced Johne's disease, allowing the transmission of M. avium subsp. paratuberculosis between animals. The objective of this study was to formulate an optimized protocol for the isolation of M. avium subsp. paratuberculosis in milk. The parameters investigated included chemical decontamination with N-acet...
متن کاملOn the functional significance of molecular variation in Symbiodinium, the symbiotic algae of Cnidaria: photosynthetic response to irradiance
The photosynthetic response to irradiance (PI response) of dinoflagellate algae of the genus Symbiodinium was quantified immediately after isolation from symbiosis with 9 species of corals and allied taxa on Bermuda. Significant variation in PI parameters was identified among the algal isolates, but no consistent differences between representatives of different Symbiodinium phylotypes were obta...
